چکیده

The ability to quantify cholesterol and/or cholesteryl ester deposits in vivo noninvasively, a goal not yet achieved, could greatly facilitate new discoveries in atherosclerosis progression and regression. In this issue of iJACC, Duivenvoorden et al. (1) report progress in a H-magnetic resonance (MR) spectroscopic technique that samples an approximate 5-mm cubic voxel in the region of the carotid artery in humans. Using cholesteryl linoleate and triolein as representative standards for cholesteryl ester and triglyceride, respectively, they suggest that the ratio of cholesteryl ester to triglyceride can be quantified and correlated with plaque tissue volume versus perivascular tissue volume in the voxel of interest. However, only one-half of the MR spectra obtained were of adequate quality for analysis.

Detection of liquid phase cholesteryl ester in carotid atherosclerosis by 1H-MR spectroscopy in humans.

OBJECTIVES This study hypothesized that (1)H magnetic resonance spectroscopy ((1)H-MRS) can identify carotid plaque cholesteryl ester in vivo in humans. BACKGROUND Liquid phase cholesteryl ester comprises a major fraction of atherosclerotic plaque, and its abundance is associated with plaque rupture and atherothrombosis. A noninvasive imaging technique to detect liquid cholesteryl ester that ...

Hepatic origin of cholesteryl oleate in coronary artery atherosclerosis in African green monkeys. Enrichment by dietary monounsaturated fat.

Relationships among plasma lipoprotein cholesterol, cholesterol secretion by the isolated, perfused liver, and coronary artery atherosclerosis were examined in African green monkeys fed diets containing cholesterol and 35% of calories as fat enriched in polyunsaturated, monounsaturated, or saturated fatty acids. The livers of animals fed monounsaturated fat had significantly higher cholesteryl ...

Role of acid lipase in cholesteryl ester accumulation during atherogenesis. Correlation of enzyme activity with acid lipase-containing macrophages in rabbit and human lesions.

Purified acid lipase was previously shown to hydrolyze the artificial substrate, alpha-naphthyl palmitate, as well as triglycerides and cholesteryl esters and to form cholesteryl esters. To determine to what extent these activities are associated with acid lipase-containing cells in atherosclerotic plaques, we examined rabbit aortas at different stages of experimental lesion induction and human...

Beta-VLDL in hepatic lipase deficiency induces apoE-mediated cholesterol ester accumulation in macrophages.

Hepatic lipase-deficient subjects in the Ontario kindred are compound heterozygotes for hepatic lipase mutations (Ser267-->Phe and Thr383-->Met). Cholesteryl ester-rich beta-very-low-density lipoprotein (beta-VLDL) accumulates in plasma and such subjects have premature atherosclerosis. To determine a possible mechanism, we hypothesized that hepatic lipase-deficient beta-VLDL, homozygous for apo...

)3-VLDL in Hepatic Lipase Deficiency Induces ApoE-Mediated Cholesterol Ester Accumulation in Macrophages

Hepatic lipase-deficient subjects in the Ontario kindred are compound heterozygotes for hepatic lipase mutations (Ser^-^Phe and Thr^^Met). Cholesteryl ester-rich 0-very-low-density lipoprotein (/3VLDL) accumulates in plasma and such subjects have premature atherosclerosis. To determine a possible mechanism, we hypothesized that hepatic lipase-deficient 0-VLDL, homozygous for apolipoprotein (apo...
